Stability depends heavily on physical state. A lyophilized powder kept dry, desiccated, and shielded from light typically holds its integrity for months to years at minus twenty degrees Celsius, and longer at minus eighty. Once dissolved, the peptide becomes far more vulnerable, since peptide bond hydrolysis, oxidation of susceptible residues, and aggregation all proceed faster in solution. Buffers near neutral pH are generally gentler than strongly acidic or alkaline conditions. Repeated freeze-thaw cycles and exposure to air-liquid interfaces during vigorous mixing cause losses that are easy to overlook.
Verification matters because research peptides vary widely in quality. A certificate of analysis is only as reliable as the method behind it, and a single chromatographic trace reveals little about counter-ions, residual solvents, or water content. Independent laboratories commonly pair mass confirmation with chromatographic purity and, where relevant, quantify water along with acetate or trifluoroacetate content. Reported purity figures are not standardized across suppliers, so a stated value such as ninety-eight percent is not directly comparable unless the analytical method, column, and detection wavelength accompany it.
Reconstitution is typically performed with sterile water or bacteriostatic water, added slowly against the vial wall. The resulting solution should be clear and colorless; cloudiness or visible particles suggest a problem with the material or the diluent. Once in solution, the peptide is less stable than the dry powder. Refrigerated storage at two to eight degrees Celsius is common for short-term holding, while freezing aliquots is described for longer periods.
Purity is most often assessed by reversed-phase high-performance liquid chromatography, reported as a percentage of total peak area. Identity is confirmed by mass spectrometry, which yields a molecular ion consistent with the expected sequence. Amino acid analysis and peptide mapping provide additional characterization. Reported purity values are method-dependent, so figures from different laboratories are not always directly comparable without details of column, gradient, and detection wavelength.

Four amino acid substitutions separate the modified backbone from the parent GRF(1-29) sequence. These changes reduce recognition by dipeptidyl peptidase IV and related proteases, extending the interval before degradation. Development work in this area sought longer-acting GHRH analogs for endocrine investigation. Published descriptions treat the substitution set as a defining property of the core sequence, while the albumin-binding linker is described separately as an optional addition to that same backbone.
CJC-1295 is a synthetic peptide analog of growth hormone-releasing hormone, constructed on the 29-amino-acid fragment designated GRF(1-29). The name began as an internal development code during the 1990s and later spread through research supply catalogs and discussion forums. The molecule does not occur in nature; its sequence is engineered rather than isolated from tissue. Two related compounds are sold under this single label, and they differ by one appended chemical group that strongly influences how long the peptide remains in circulation.
The peptide backbone corresponds to GRF(1-29), the first 29 residues of native growth hormone-releasing hormone, which retains most of the receptor-activating activity of the full-length molecule. Four substitutions distinguish the analog from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes slow cleavage by dipeptidyl peptidase IV, the enzyme that degrades native hormone in plasma within minutes. The outcome is improved enzymatic stability combined with a still brief residence time when no additional modification is present.
Naming in this area is inconsistent, and readers should treat product labels with care. In much of the literature and in vendor catalogs, the unqualified term refers to the albumin-binding version, while the version lacking the DAC group appears as modified GRF(1-29), mod GRF(1-29), or the same name with a without-DAC qualifier. Because one abbreviation has been applied to both materials, the only reliable way to identify a sample is to check the stated sequence and the presence of the linker.
CJC-1295 is the name used for a synthetic peptide modeled on growth hormone-releasing hormone, the hypothalamic signal that prompts the pituitary to release growth hormone. The compound was described by a Canadian drug discovery company in the mid-2000s as a long-acting research tool. Two closely related molecules share the name in practice: one carries a drug affinity complex, or DAC, group, and one does not. The distinction matters because the two behave differently in circulation.
Laboratory handling centers on minimizing exposure to water, heat and oxygen before use. Working solutions are typically prepared in sterile water or a mild buffer, and any residual particulate matter is removed by filtration. When the powder dissolves slowly, a small proportion of acetonitrile or dilute acetic acid is sometimes added as a co-solvent. Containers are kept sealed and desiccated between uses. Records of lot number, reconstitution date and storage conditions support later comparison of results across experiments.
Identity and purity are established with reversed phase high performance liquid chromatography coupled to mass spectrometry. The chromatographic step separates the target peptide from truncated sequences and deletion products, while the mass measurement confirms the expected molecular weight to within a fraction of a dalton. Because the two common variants differ by the presence of the linker, mass alone can distinguish them in the unconjugated state. Amino acid analysis and peptide mapping are used when sequence level confirmation is required.
Paul, and Stephen Walt concede that traditional balancing is not occurring, but argue nevertheless that rivals to the US are engaging in 'soft balancing.'More recent scholarly work has engaged the debate on soft balancing. Kai He suggested a new analytical framework, a negative balancing model, to explain why states do not form alliances or conduct arms races to balance against power or threats as they may have done in the past. He describes negative balancing as any strategy or diplomatic efforts aimed to undermine a rival's power. In contrast, positive balancing is actions or policies designed to strengthen a state's own power in world politics.
While most early submarines used a direct mechanical connection between the combustion engine and the propeller, an alternative solution was considered as well as implemented at a very early stage. That solution consists in first converting the work of the combustion engine into electric energy via a dedicated generator. This energy is then used to drive the propeller via the electric motor and, to the extent required, for charging the batteries. In this configuration, the electric motor is thus responsible for driving the propeller at all times, regardless of whether air is available so that the combustion engine can also be used or not. Among the pioneers of this alternative solution was the very first submarine of the Swedish Navy, HSwMS Hajen (later renamed Ub no 1), launched in 1904. While its design was generally inspired by the first submarine commissioned by the US Navy, USS Holland, it deviated from the latter in at least three significant ways: by adding a periscope, by replacing the gasoline engine by a semidiesel engine (a hot-bulb engine primarily meant to be fueled by kerosene, later replaced by a true diesel engine) and by severing the mechanical link between the combustion engine and the propeller by instead letting the former drive a dedicated generator. By so doing, it took three significant steps toward what was eventually to become the dominant technology for conventional (i.e., non-nuclear) submarines.
== Modifications == As Black Mesa is built on the Source engine, it itself is also moddable with support for the Steam Workshop, and several projects have been started to create versions of Half-Life mods and expansions within Black Mesa.
Alongside dame school, Thomas also took private lessons from Gwen James, an elocution teacher who had studied at drama school in London, winning several major prizes. She also taught "Dramatic Art" and "Voice Production", and would often help cast members of the Swansea Little Theatre (see below) with the parts they were playing. Thomas's parents' storytelling and dramatic talents, as well as their theatre-going interests, could also have contributed to the young Thomas's interest in performance. In October 1925, Thomas enrolled at Swansea Grammar School for boys, in Mount Pleasant, where his father taught English. There are several accounts by his teachers and fellow pupils of Thomas's time at grammar school. In June 1928, Thomas won the school's mile race, held at St. Helen's Ground; he carried a newspaper photograph of his victory with him until his death. As a pupil he shied away from the school curriculum, preferring his own choice of reading, drama activities and contributing to the school's magazine of which he became editor. Examples of plagiarised poems he published under his own name in the magazine, and in some cases more widely, have been noted by his biographers. Subsequent research has revealed the extent and virtuosity of Thomas's plagiarism, including having a poem published in the nationally read Boy's Own Paper that had been published in the same magazine 15 years before. During his final school years he began writing his distinctively original poetry in notebooks; the first poem, dated 27 April (1930), is entitled "Osiris, come to Isis".
=== Suppression of the pro-Alid groups === Mu'awiya died in 680 and was succeeded by his son Yazid I. Mu'awiya's designation of his son was an unprecedented act and shocked many in the Muslim community, particularly the Arab nobility of Kufa. They long sympathized with Caliph Ali, Mu'awiya's former rival, and Ali's family. One of Ali's sons, Husayn dispatched his cousin Muslim ibn Aqil to Kufa to set the stage for Husayn's accession to the caliphate. Ibn Aqil garnered significant support and was hosted by a prominent pro-Alid nobleman. Ubayd Allah became aware of Ibn Aqil's activities, prompting the latter to launch a premature assault against the governor. Ubayd Allah was holed up in his palace, but thirty men from his shurta (security forces) fended off Ibn Aqil's partisans, while he persuaded many Kufan noblemen to back him against Ibn Aqil, who was abandoned by his supporters and slain on 10 September 680. Husayn had already been en route to Kufa from Medina when he received news of Ibn Aqil's execution. Ubayd Allah was prepared for Husayn's arrival and sent troops to intercept him. They prevented Husayn and his small retinue from reaching the watered areas of the province. The two sides negotiated for weeks, but Ubayd Allah refused Husayn entry into Kufa or return to Arabia while Husayn refused to recognize Yazid's caliphate. In the end, a short battle was fought at Karbala on 10 October 680, in which Husayn and nearly all of his partisans were slain.

Since the species is only found in remnant populations in the Wallacean Island, studies have examined the traits and factors that led to these certain populations' surprising persistence. A comprehensive examination of the species survival suggested that rather than being a single attribute, but the a combination including ectotherm traits, varanid biology, habitat, and anthropogenic factors.
The three substrates of this enzyme are crotonic acid, reduced nicotinamide adenine dinucleotide (NADH), and a proton. Its products are butyric acid and oxidised NAD+. This enzyme belongs to the family of oxidoreductases, specifically those acting on the CH-CH group of donor with NAD+ or NADP+ as acceptor. The systematic name of this enzyme class is butanoate:NAD+ Delta2-oxidoreductase. This enzyme is also called enoate reductase. This enzyme participates in phenylalanine metabolism. It has cofactors: flavin adenine dinucleotide and iron–sulfur protein.
=== Complications === Complications of smallpox arise most commonly in the respiratory system and range from simple bronchitis to fatal pneumonia. Respiratory complications tend to develop on about the eighth day of the illness and can be either viral or bacterial in origin. Secondary bacterial infection of the skin is a relatively uncommon complication of smallpox. When this occurs, the fever usually remains elevated. Other complications include encephalitis (1 in 500 patients), which is more common in adults and may cause temporary disability; permanent pitted scars, most notably on the face; and complications involving the eyes (2% of all cases). Pustules can form on the eyelid, conjunctiva, and cornea, leading to complications such as conjunctivitis, keratitis, corneal ulcer, iritis, iridocyclitis, and atrophy of the optic nerve. Blindness results in approximately 35–40% of eyes affected with keratitis and corneal ulcer. Hemorrhagic smallpox can cause subconjunctival and retinal hemorrhages. In 2–5% of young children with smallpox, virions reach the joints and bone, causing osteomyelitis variolosa. Bony lesions are symmetrical, most common in the elbows, legs, and characteristically cause separation of the epiphysis and marked periosteal reactions. Swollen joints limit movement, and arthritis may lead to limb deformities, ankylosis, malformed bones, flail joints, and stubby fingers. Between 65 and 80% of survivors conserve indelible white scars.
== Operations == Dutch Bros is headquartered in Tempe, Arizona. It had been based in Grants Pass, Oregon, since its 1992 founding, but began moving some corporate offices to Arizona in early 2024 and in June 2025 announced that it was relocating its headquarters there. The company is majority-owned by Travis Boersma, who holds the title of executive chairman, and Christine Barone is its president and CEO. In September 2021, Dutch Bros became a publicly traded company, selling 21 million shares for a total of $484 million. As of December 2023, the company employed approximately 24,000 people, and as of August 2024, it had 912 stores. In 2024, Dutch Bros opened a distribution and roasting center in Melissa, Texas. The center is a 65,000-square-foot plant located at the intersection of U.S. 75 and State Highway 121. In January 2026, it was announced that Dutch Bros had agreed to acquire Clutch Coffee Bar, a regional drive-through coffee chain with 20 locations across North and South Carolina. The financial terms were not disclosed, and Dutch Bros stated that the acquired locations would be closed, renovated, and reopened under its own brand. On August 5, 2026, the company announced it would be acquiring 65 locations from the chain Salad and Go (which permanently closed all its locations the same day) across Arizona, Nevada, Oklahoma and Texas.

Mass spectrometry establishes whether the observed molecular weight matches the calculated sequence mass. Chromatographic retention and fragment mapping add further confidence about sequence and composition. A single technique alone is rarely treated as sufficient evidence of identity.
Dry powder is markedly more stable than solution and is usually held frozen and desiccated. Room temperature exposure over days degrades it more slowly than most people assume, but long-term integrity favors freezer storage. Light protection is recommended regardless of temperature.
Methods, columns, gradients, and detection wavelengths are not standardized across laboratories. Some figures describe only the main peak area at one wavelength and ignore water, salts, or counter-ions. Without stated conditions, comparing reported percentages across sources is unreliable.
Cool, dark, and dry conditions are standard, with storage at minus twenty degrees Celsius or below. Desiccant and sealed vials limit moisture uptake. Repeated warming and cooling of the container is generally avoided.
