storage raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

Two forms circulate in research settings and are frequently confused. One carries the drug affinity complex and is often written as CJC-1295 with DAC; the other lacks that group and is usually called modified GRF(1-29). The two share the same core sequence but differ sharply in how long they persist in blood. Products labelled only as CJC-1295 normally refer to the version carrying the complex. Documentation that omits the distinction leaves the intended molecule ambiguous.
CJC-1295 is a synthetic peptide built as a long-acting analogue of growth hormone-releasing hormone. Its backbone matches the first twenty-nine residues of the natural human hormone, with four amino acid substitutions added to slow enzymatic breakdown. A reactive maleimide group, commonly termed the drug affinity complex, allows the peptide to attach to circulating albumin after administration. That albumin attachment keeps the molecule in the bloodstream for an extended period instead of being cleared within minutes.
Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone, built on the 29-amino-acid GHRH(1-29) fragment. Four substitutions distinguish it from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes reduce enzymatic cleavage and extend the peptide's persistence in circulation. The compound is discussed in two forms, one carrying a drug affinity complex and one without it.

Four amino acid substitutions separate the modified backbone from the parent GRF(1-29) sequence. These changes reduce recognition by dipeptidyl peptidase IV and related proteases, extending the interval before degradation. Development work in this area sought longer-acting GHRH analogs for endocrine investigation. Published descriptions treat the substitution set as a defining property of the core sequence, while the albumin-binding linker is described separately as an optional addition to that same backbone.
CJC-1295 is a synthetic peptide analog of growth hormone-releasing hormone, constructed on the 29-amino-acid fragment designated GRF(1-29). The name began as an internal development code during the 1990s and later spread through research supply catalogs and discussion forums. The molecule does not occur in nature; its sequence is engineered rather than isolated from tissue. Two related compounds are sold under this single label, and they differ by one appended chemical group that strongly influences how long the peptide remains in circulation.
The dividing feature between the two forms is a maleimide-based drug affinity complex, abbreviated DAC. In the DAC-bearing version, a linker attaches the peptide to serum albumin after administration, and that association slows removal from plasma. The version lacking DAC appears in catalogs as MOD GRF(1-29) or tetrasubstituted GRF(1-29). Because informal writing treats both as one item, comparisons drawn from such sources routinely blend measurements taken from two molecules with substantially different behavior.
Both forms act at the pituitary receptor for growth hormone-releasing hormone and increase growth hormone output, which in turn raises insulin-like growth factor 1. A long-acting analog produces sustained rather than pulsatile stimulation, and the physiological consequences of that pattern are not fully settled. Published human data on the extended form remain limited, and much of what circulates in discussion traces to early company reports rather than independent replication. How sustained exposure affects normal feedback remains an open question.
The distinguishing feature of the DAC form is a maleimide-containing group that reacts with the free thiol of cysteine-34 on human serum albumin. This reaction forms a covalent bond without enzymatic assistance, and it takes place after the peptide enters the bloodstream. Because albumin is abundant and long-lived, the attached peptide is carried through circulation far longer than an unmodified fragment would survive. The chemistry is a deliberate pharmacokinetic strategy rather than a change to receptor activity.
Enzymatic protection is a separate mechanism from plasma protein binding. The four substitutions in the backbone reduce recognition by dipeptidyl peptidase IV, which normally cleaves the natural hormone within minutes. Without the reactive group, this resistance still yields only a short window of activity, generally reported in the range of tens of minutes. With it, reported half-lives in early human work extended to several days. The size of that gap is the main practical distinction between the two materials.
Reverse-phase high-performance liquid chromatography is the standard tool for purity assessment. The technique separates the target peptide from truncated or modified byproducts. Mass spectrometry confirms molecular weight and supports sequence verification. Electrospray ionization and matrix-assisted laser desorption are both used. Amino acid analysis provides an independent check on composition. Purity values are commonly reported as area percentage from the chromatogram. Residual trifluoroacetate and water content are also measured in many quality programs.
Batch-to-batch consistency depends on solid-phase peptide synthesis and subsequent purification. Coupling efficiency, resin choice, and cleavage conditions all affect the final profile. Counter-ion content and moisture can shift the apparent mass of a batch. Documentation typically includes a certificate of analysis with chromatograms and spectra. Independent verification by a second laboratory is sometimes requested. Whether a given certificate reflects the actual vial contents depends on chain of custody. Analytical methods themselves carry uncertainty that should be stated alongside results.
Lyophilized material is typically stored at minus twenty degrees Celsius or lower. Keeping the vial dry and protected from light preserves peptide integrity. Repeated freeze-thaw cycles can cause aggregation or loss of activity. Once dissolved, solutions are generally kept at two to eight degrees Celsius. Stability data for reconstituted solutions vary, and long-term behavior is not fully established. Working aliquots reduce the number of times a stock container is opened.
Studies in this area generally track growth hormone pulses, insulin-like growth factor 1 concentrations, and occasionally body composition endpoints. Most published human data come from early, small trials, and questions about long-term effects remain open. Whether repeated exposure alters pituitary responsiveness over time is not settled. Analytical work relies on immunoassays for the hormones and on mass spectrometry for the peptide itself, because the two measurements answer different questions.
Binding of the peptide to the growth hormone-releasing hormone receptor on pituitary somatotrophs triggers a G protein coupled cascade that raises cyclic AMP and opens calcium channels. The result is greater secretion of growth hormone into the bloodstream. Because the peptide acts at the same receptor as the natural hypothalamic hormone, its effect is amplified pulse size rather than an entirely separate release pathway. Receptor binding alone does not determine the response, since somatostatin tone and other inputs modulate the final output.
==== Statistical power ==== In any clinical trial, the number of subjects, also called the sample size, has a large impact on the ability to reliably detect and measure the effects of the intervention. This ability is described as its "power", which must be calculated before initiating a study to figure out if the study is worth its costs. In general, a larger sample size increases the statistical power, also the cost. The statistical power estimates the ability of a trial to detect a difference of a particular size (or larger) between the treatment and control groups. For example, a trial of a lipid-lowering drug versus placebo with 100 patients in each group might have a power of 0.90 to detect a difference between placebo and trial groups receiving dosage of 10 mg/dL or more, but only 0.70 to detect a difference of 6 mg/dL.
== Society and culture == Multiple black market sellers of the peptide emerged before approval of the Lilly drug, leading to multiple lawsuits with Lilly as the plaintiff. Eli Lilly sued six entities: Lone Star Peptide Co., Aesthetic Envy Cosmetic Centers, Striker Pharmacy, Texas Peptides, Astra LLC (Astra Peptides), and Legendary Peptides.
=== Surgery === On 12 June 1831, Dupuytren performed a surgical procedure on a person with contracture of the fourth and fifth digits who had been previously told by other surgeons that the only remedy was cutting the flexor tendons. He described the condition and the operation in The Lancet in 1834 after presenting it in 1833, and posthumously in 1836 in a French publication by Hôtel-Dieu de Paris. The procedure he described was a minimally invasive needle procedure. Because of high recurrence rates, new surgical techniques were introduced, such as fasciectomy and then dermofasciectomy. Most of the diseased tissue is removed with these procedures.Recurrence rates are low. For some individuals, the partial insertion of "K-wires" into either the DIP or PIP joint of the affected digit for a period of a least 21 days to fuse the joint is the only way to halt the disease's progress. After removal of the wires, the joint is fixed into flexion, which is considered preferable to fusion at extension. Research using large datasets in the UK has shown surgery to be safe and effective. When surgery needs to be repeated, however, the research suggests there are higher risks of serious complications such as finger amputation. Amputation of fingers may be needed for severe or recurrent cases or after surgical complications.
6 HPO2−4 + 2 H2O + 10 Ca2+ ⇌ Ca10(PO4)6(OH)2 + 8 H+ In a closed system as mineral precipitates, acid accumulates, rapidly lowering the pH and stopping further precipitation. Cartilage presents no barrier to diffusion and acid therefore diffuses away, allowing precipitation to continue. In the osteon, where matrix is separated from extracellular fluid by tight junctions, this cannot occur. In the controlled, sealed compartment, removing H+ drives precipitation under a wide variety of extracellular conditions, as long as calcium and phosphate are available in the matrix compartment. The mechanism by which acid transits the barrier layer remains uncertain. Osteoblasts have capacity for Na+/H+ exchange via the redundant Na/H exchangers, NHE1 and NHE6. This H+ exchange is a major element in acid removal, although the mechanism by which H+ is transported from the matrix space into the barrier osteoblast is not known. In bone removal, a reverse transport mechanism uses acid delivered to the mineralized matrix to drive hydroxyapatite into solution.
Another synonymous mutation (c.3156C>T) was proposed to introduce a cryptic donor site, resulting in aberrant splicing, a theory which subsequently was confirmed in several families with ERED from different countries.

==== Deposition ==== The extracellular matrix of bone is laid down by osteoblasts, which secrete both collagen and ground substance. These cells synthesise collagen alpha polypeptide chains and then secrete collagen molecules. The collagen molecules associate with their neighbors and crosslink via lysyl oxidase to form collagen fibrils. At this stage, they are not yet mineralized, and this zone of unmineralized collagen fibrils is called "osteoid". Around and inside collagen fibrils calcium and phosphate eventually precipitate within days to weeks becoming then fully mineralized bone with an overall carbonate substituted hydroxyapatite inorganic phase. In order to mineralise the bone, the osteoblasts secrete alkaline phosphatase, some of which is carried by vesicles. This cleaves the inhibitory pyrophosphate and simultaneously generates free phosphate ions for mineralization, acting as the foci for calcium and phosphate deposition. Vesicles may initiate some of the early mineralization events by rupturing and acting as a centre for crystals to grow on. Bone mineral may be formed from globular and plate structures, and via initially amorphous phases.
Calcium (20Ca) has 26 known isotopes, ranging from 35Ca to 60Ca. There are five stable isotopes (40Ca, 42Ca, 43Ca, 44Ca and 46Ca), plus one isotope (48Ca) with such a long half-life that it is for all practical purposes stable. Two isotopes of calcium, 40Ca and 48Ca, are doubly-magic, making them particularly stable nucliides. Calcium also has a cosmogenic isotope, 41Ca, with half-life 99,400 years. Unlike cosmogenic isotopes produced in the air, 41Ca is produced by neutron activation of solid 40Ca in rock and soil. Most of its production is in the upper meter of the soil column, where the cosmogenic neutron flux is still strong enough. Calcium-41 is also produced during stellar nucleosynthesis and decays to 41K, which can be used to understand formation and mixing between solar system reservoirs . The most stable artificial isotopes are 45Ca with half-life 162.61 days and 47Ca with half-life 4.536 days. All other artificial calcium isotopes have half-lives of minutes or less. 40Ca comprises about 97% of natural calcium and is mainly created by nucleosynthesis in stars (alpha process). Like 40Ar, however, some 40Ca is radiogenic, created by radioactive decay of 40K. While K–Ar dating has been used extensively in the geological sciences, the prevalence of 40Ca in nature initially impeded the proliferation of K-Ca dating in early studies, with only a handful of studies in the 20th century.
An Iranian ballistic missile strike damaged an Israeli drone factory in Petah Tikva. On the same day, Iran's army chief, Amir Hatami, stated that if US forces invade the country, "not a single person" among the invaders will survive.
(2026) reconstruct the diet of Miocene rodents and South American native ungulates from the La Venta site (Colombia) on the basis of stable isotope analyses of tooth enamel, reporting evidence of C3-dominated diets of the studied mammals, but also evidence of ecological differentiation. Casanovas-Vilar et al. (2026) interpret the small mammal assemblage from the Vallès-Penedès Basin (Spain) as consistent with a climate during the Miocene that was transitional between humid subtropical and Mediterranean types, finding no evidence of significant climate changes at the times of major faunal turnovers. Martín-Perea et al. (2026) study the taphonomy of late Miocene mammalian assemblages from the Daidian and Dongmen sites (China), and interpret the studied assemblages as resulting from gradual accumulation through attritional mortality. Evidence from fossil data from low latitudes in Mexico, indicating that the dispersal of North American mammals into South America during the Great American Interchange was preceded and primed by their accumulation in Mexico beginning 10 million years ago, is presented by Tseng et al. (2026).
==== Chemical secretion ==== Recognition of MAMPs by PRRs can activate tissue resident macrophages to secrete proinflammatory cytokines that recruit other immune cells. Among the PRRs, TLRs play a major role in signal transduction leading to cytokine production. The binding of MAMPs to TLR triggers a series of downstream events that eventually activates transcription factor NF-κB and results in transcription of the genes for several proinflammatory cytokines, including IL-1β, IL-6, TNF-α, IL-12B, and type I interferons such as IFN-α and IFN-β. Systemically, IL-1β, IL-6, and TNF-α induce fever and initiate the acute phase response in which the liver secretes acute phase proteins. Locally, IL-1β and TNF-α cause vasodilation, where the gaps between blood vessel epithelial cells widen, and upregulation of cell surface adhesion molecules on epithelial cells to induce leukocyte extravasation. Additionally, activated macrophages have been found to have delayed synthesis of prostaglandins (PGs) which are important mediators of inflammation and pain. Among the PGs, anti-inflammatory PGE2 and pro-inflammatory PGD2 increase the most after activation, with PGE2 increasing expression of IL-10 and inhibiting production of TNFs via the COX-2 pathway. Neutrophils are among the first immune cells recruited by macrophages to exit the blood via extravasation and arrive at the infection site. Macrophages secrete many chemokines such as CXCL1, CXCL2, and CXCL8 (IL-8) that attract neutrophils to the site of infection.

(SC) subcutaneous (most applicable to real bites): 0.32 mg/kg, 0.28 mg/kg. (IV) intravenous: 0.25 mg/kg, 0.011 mg/kg. (IP) intraperitoneal: 0.30 mg/kg (average), 0.941 mg/kg. 0.05 mg/kg (the last quote does not make it clear if it is either intravenous or intraperitoneal). It is estimated that only 10 to 15 mg will kill a human adult; however, its bites deliver about 120 mg of venom on average, although they may deliver up to 400 mg of venom in a single bite. Severe neurotoxicity invariably ensues rapidly. This is because of several factors, including the high potency of the venom, and the unusual composition of synergistic toxins contained within the venom, which bring on severe symptoms of envenomation and death much faster than any other snake venom. In addition, the snake itself possesses the most advanced, evolved, and efficient venom delivery apparatus among all venomous snakes, and the most advanced and evolved dentition of all elapids. Black mambas possess the longest fangs of any elapid, which average around 13.1 millimeters (0.52 in), but may grow as long as 22 millimeters (0.87 in). Another feature that distinguishes the dentition and venom delivery apparatus of this species from all other elapids, as well as other species of venomous snake including those of the family Viperidae, is that the fangs are positioned well forward at the most anterior position possible in its mouth – right up in the front of the upper jaw.
=== Personal guard of Prince Menelik === The Mehal Sefari's earlier roles were providing security for young Prince Menelik under the leadership of the later Dejazmach Germame. Upon the death of Atse Tewodros, 3 men – Wagshum Gobeze of Welo, Kassa Mircha of Tigre and Menelik of Shoa – were declared Atse. Kassa marched on Welo, defeated and imprisoned Gobeze, he marched south to Shewa to face Menelik who had gathered his forces and awaited him. Both rulers assumptive, Yohannes by virtue of arms left for him by the British and Menelik by blood sought reconciliation, Menelik agreeing to accept Yohannes as his Suzerain, much to the relief of the latter whose small, though well-armed forces were no match for the Shewan Army. Menelik's Army, though with fewer artillery pieces, had superiority in men, equipment and mounted cavalry. It would also have been fighting on home turf. Yohannes' spy sent to observe the Shoans is said to have come back to tell the Emperor "I thought clouds had descended on the ground, but it was the Shoans and their multitudes in their tents." Atse Menelik took this opportunity to continue to arm and train his men, dispatching men to the South and West in poses of providing security for the Emperor of Ethiopia and an elite infantry division.
Mated females then seek a suitable place to begin a colony. There, they break off their wings using their tibial spurs and begin to lay and care for eggs. The females can selectively fertilise future eggs with the sperm stored to produce diploid workers or lay unfertilized haploid eggs to produce drones. The first workers to hatch, known as nanitics, are weaker and smaller than later workers but they begin to serve the colony immediately. They enlarge the nest, forage for food, and care for the other eggs. Species that have multiple queens may have a queen leaving the nest along with some workers to found a colony at a new site, a process akin to swarming in honeybees.
==== MHC class II molecule stabilization - chaperonal function ==== The low pH of lysosomes could cause denaturation or proteolysis of MHC class II molecules. HLA-DM binding to MHC stabilizes and protects from degradation, by covering hydrophobic surfaces. Antigen degradation could also ensue, resulting in an inability to bind to the peptide-binding groove. Thus, HLA-DM is needed to protect proteins against the lysosomal environment.
Mahathir's political views have shifted during his lengthy career. During the 1980s and 1990s, he was a supporter of third-worldism, while during other periods he has been a proponent of "Asian values" and globalisation. A Muslim thinker, he holds Islamic political views. In 2002, he characterised himself as an Islamic fundamentalist. Mahathir is generally respected in developing and Islamic countries, particularly due to his oversight of Malaysia's economic growth and his support of liberal Muslim values. Mahathir has been described as a proponent of Malay nationalism. In The Malay Dilemma, he argued that the Malay race had been marginalised, and voiced his support for affirmative action policies for them. Upon his first resignation, he expressed his disappointment at the progress made towards his "principle task" of supporting the Malay race. In 2021, Mahathir said he did not believe in Ketuanan Melayu, calling it a "fantasy", and said instead that he believed in the concept of "Bangsa Malaysia". He has been described as anti-royalist by the left-wing French newspaper Libération, owing to his efforts to oppose immunity for members of Malaysia's monarchies. Mahathir is a vocal critic of neoliberalism and the Western world. In 2011, Mahathir suggested that the September 11 attacks might have been staged by the United States government. Mahathir condemned the Universal Declaration of Human Rights in 1997, suggesting it be revised to place greater importance on economic growth over civil liberties.

It is a synthetic peptide analogue of growth hormone-releasing hormone. Four substitutions in its sequence make it more resistant to enzymatic degradation than the natural hormone. In the version carrying a drug affinity complex, the peptide binds albumin and remains in circulation for days.
No regulatory agency has approved CJC-1295 for clinical use. Human trials were conducted in the 2000s, but the development programme was discontinued before any marketing application succeeded. Material sold today is offered as a research chemical, and its purity depends on the supplier.
Sermorelin is an unmodified fragment of growth hormone-releasing hormone and is cleared quickly. CJC-1295 contains substitutions that resist breakdown, and the form with a drug affinity complex persists much longer. Both act at the same receptor but differ substantially in duration of action.
The two share the same 29-amino-acid backbone. CJC-1295 carries an additional albumin-binding moiety that markedly extends its residence time in circulation, while the form without that moiety clears faster. Literature sometimes applies the name loosely to either variant.
